Most current experimental models in cancer biology involve the use of cancer cell lines. These are cell lines derived from human tumor tissue and “adapted” to growth in in vitro culture. This process results in subpopulations of tumor derived cells that have been “selected” for growth in conditions that are very different from the tumor microenvironment.
Xenograft tumors generated in mice from engrafted cancer cell lines have been the standard for pre-clinical drug development studies. However, it has become increasingly clear that cultured cancer cells are highly divergent from the original tissue. This divergence between “laboratory” cancer cells and the cells in human tumors has significantly hampered the successful translation of drug development studies from the bench to the clinic.
PDTX tumor models recapitulate the pathological features of the original tumor tissue. Numerous studies have demonstrated that PDTX tumors retain histological features, biomarker expression, genetic and genomic profiles of the engrafted tumor. Patient-derived xenografts are a powerful tool for the measurement of compound activity, as the engrafted tumor growth rate can be easily measured. These complex studies require selection of the correct animal model to go with the tumor type, as well as selection of administration method and expert analysis of tumor growth. We have years of experience in in-house xenotransplantation studies and provide patient derived xenograft services at Altogen Labs, allowing us to offer optimal experiment plans and execution.
Patient derived xenograft services are commercially available (please request study quotation here) and provides establishment of PDTX models, complete characterization of newly established models and use of existing PDTX models. All animal handling at our Altogen Labs facility is IACUC-regulated and complies with all current Good Laboratory Practices (GLPs). Our clients receive detailed procedures, data and animal health reports. In addition to standard services, we also offer tox and biodistridution studies, histology, tissue collection, biomarker analysis, RNA isolation and gene expression analysis.
